ELISA中的试剂－免疫吸附剂 临床检验中一般采用商品盒进行测定。ELISA中有三个必要的：免疫吸附剂、结合物和酶的底物。完整的ELISA盒包含以下各组分：（1）已包被抗原或的固相载体（免疫吸附剂）；（2）酶标记的抗原或（结合物）；（3）酶的底物；（4）阴性对照品和阳性对照品（定性测定中），参考标准品和控制血清（定量测定中）；（5）结合物及标本的稀释液；（6）洗涤液；（7）酶反应终止液。3.1 免疫吸附剂已包被抗原或的固相载体在低温（2~8℃）干燥的条件下一般可保存6个月。有些不完整的试盒，仅供应包被用抗原或抗体，检测人员需自行包被。以下简述固相载体和包被过程。3.1.1 固相载体固相载体在ELISA测定过程中作为吸附剂和容器，不参与化学反应。可作ELISA中载体的材料很多，最常用的是聚苯乙烯。聚苯乙烯具有较强的吸附蛋白质的性能，抗体或蛋白质抗原吸附其上后仍保留原来的免疫学活性，加之它的价格低廉，所以被普遍采用。聚苯乙烯为塑料，......阅读全文
