怎样正确进行ELISA操作一、临床标本的收集和保存 用于ELISA测定的临床标本最为常用的是血清（浆），有时因为特定的检测目的，也用到唾液、脑脊液、尿液、粪便等标本。目前临床上使用血清标本测定的标志物一般有传染性病原体的抗原和抗体、肿瘤标志物、激素、特种蛋白、细胞因子和治疗药物等。对用于激素和治疗药物测定的血清标本的收集，要注意收集时间甚或体位有可能会对测定结果产生影响。如可的松在早晨4～6点之间，会有一峰值出现：生长激素、促黄体激素（LH）和促卵泡激素（FSH）均以阵发性方式释放，因此，在测定此类激素时，有必要在密切相连的时间间隔内采取数份血样本，以其中间值为测定值。又如当从卧位变为站立位时，血清中肾素活性将出现明显增高。再如治疗药物的检测，应根据药代动力学选择服药后的最适时间抽血检测。用于传染性病原体的抗原和抗体、肿瘤标志物和特种蛋白等的检测的血清标本的收集则没有时间和体位方面的影响，只是在处理和保存方面要考虑以下几个方......阅读全文
