免疫球蛋白提取技术(Immunoglobulinisolationtechnique)
免疫球蛋白（Immunoglobulin Ig）的含量代表着机体体液免疫的水平，并进一步代表着B细胞的功能，因此测定血清Ig含量可以推知机体的体液免疫功能和诊断某些疾病引起的Ig的过高和过低。 随着免疫学的发展和需要，免疫球蛋白的纯化和其成分的提纯成为必不可少的手段。纯化的方法很多，有单一法，但大多数采用二步法以上相结合的方法，特别是以硫酸铵提纯为基础，再经过层析柱的方法来提高免疫球蛋白及其各成分的纯度最为常用。 硫酸铵溶液能使蛋白质胶体脱水并中和其电荷而使之沉淀下来（称为盐析）。不同浓度的硫酸铵盐析蛋白成分不同，利用这一原理提取所需的免疫球蛋白成分。盐析只能粗提，为了获得纯化的免疫球蛋白成分，必须进一步采用层析的方法进行分离。 免疫球蛋白包括IgG、IgM、IgA、IgE和IgD。 一、 IgG的分离与提纯 &......阅读全文
