实验材料CD34+细胞试剂、试剂盒灭菌MSCs培养基丝裂霉素C100μg mL共培养的培养基细胞因子(干细胞因子IL-6Flt-3配体促血小板生成素)仪器、耗材6孔培养板实验步骤(a)将脐带血来源的间充质干细胞(blood-derived mesenchymal stem cells, CB-MSCs)作为饲养层细胞,用MSCs培养基重悬CB-MSCs,细胞浓度为5×104个细胞/mL。(b)将CB-MSCs接种到6孔板(C)每周两次半量更换新鲜培养基。(d)当CB-MSCs达到以上汇合时,用10μg/mL丝裂霉素c处理,37℃ 2.5 h„(e)用无血清IMDM将CB-MSCs洗两遍。(f)按1×104个/mL CD34+细胞重悬于含细胞因子的共同培养基(100 ng/mL干细胞因子,100ng/mL IL-6,50 ng/mLFlt-3配体,10 ng/mL促血小板生成素)。(g)将CD34+细胞接种到CB-MSCs饲养层细......阅读全文
